NRAS ASO-1 is an antisense oligonucleotide specifically designed to bind the mRNA of the **NRAS gene**, thereby inhibiting its expression through targeted RNA degradation. Antisense oligonucleotides (ASOs) like NRAS ASO-1 interact with complementary sequences on NRAS mRNA by Watson-Crick base pairing. The principal mechanism involves recruitment of **RNase H1**, leading to mRNA cleavage and subsequent reduction of **NRAS protein** levels[1][2][3][5][7]. This strategy is especially relevant in cancers such as NRAS-mutant melanoma, where pathologic NRAS signaling drives abnormal cell proliferation. ASOs may also utilize alternative mechanisms such as steric hindrance of mRNA function or interference with splicing, but for NRAS ASO-1, RNA degradation is primary[1][2][3][7]. Chemical modifications to the ASO backbone and sugar moieties—such as phosphorothioate or 2'-O-methoxyethyl groups—are often used to improve stability and binding affinity, but the precise chemistry of NRAS ASO-1 may require direct confirmation[1][5].
