NRDN-201 is a preclinical-stage small molecule drug candidate being developed by Neurodon Corp. for the treatment of neurodegenerative diseases, specifically Parkinson's disease and Alzheimer's disease. The drug acts as an allosteric activator of the sarco/endoplasmic reticulum Ca2+-ATPase (SERCA) enzyme, which is responsible for transporting calcium from the cytosol into the endoplasmic reticulum. By accelerating SERCA function, NRDN-201 restores cellular calcium homeostasis and relieves endoplasmic reticulum (ER) stress, addressing key pathological drivers of neuronal dysfunction and death. In Parkinson's disease, it targets calcium balance in dopaminergic neurons of the substantia nigra, while in Alzheimer's disease, it addresses calcium dyshomeostasis in hippocampal neurons to mitigate neuroinflammation, tau phosphorylation, and amyloid aggregation.
