Overview

Nrf2-siRNA is a small interfering RNA (siRNA) designed to silence the expression of the Nuclear factor erythroid 2-related factor 2 (Nrf2), a transcription factor encoded by the NFE2L2 gene. Nrf2 is a master regulator of the cellular antioxidant response and detoxification enzymes, and its overexpression in various malignancies, such as esophageal cancer, is associated with tumor growth, chemoresistance, and poor prognosis. Nrf2-siRNA functions through the RNA interference (RNAi) pathway, where it mediates the sequence-specific degradation of Nrf2 mRNA, leading to a significant reduction in Nrf2 protein levels. In preclinical studies, treatment with Nrf2-siRNA has been shown to inhibit cancer cell proliferation and induce cell cycle arrest at the G1 phase, suggesting that targeting the Nrf2 pathway may be a viable therapeutic strategy for improving outcomes in patients with Nrf2-overexpressing tumors.
