NRG1 siRNA is a small interfering RNA (siRNA) designed to silence the expression of the neuregulin-1 (NRG1) gene through RNA interference. NRG1 is a growth factor and a ligand for the ErbB3 (HER3) receptor; its binding induces ErbB3/ErbB2 heterodimerization and activates downstream survival pathways, including the PI3K/AKT signaling cascade. In preclinical research, particularly in cutaneous melanoma models that are wild-type for BRAF and NRAS, NRG1 siRNA has been utilized to demonstrate that autocrine NRG1 production drives tumor cell viability. By knocking down NRG1 expression, the siRNA reduces ErbB3 and AKT phosphorylation, thereby inhibiting tumor cell growth and survival. This approach highlights the NRG1-ErbB3-ErbB2 axis as a potential therapeutic target in specific cancer subsets.
