Overview

NRM-823 is a first-in-class, engineered T cell engager (TCE) antibody developed by Normunity for the treatment of multiple solid tumors. It is designed to redirect T cells to attack cancer cells by binding simultaneously to a highly specific, novel tumor-associated antigen expressed across various solid tumors and the CD3 receptor on T cells. This dual binding forms an immunological synapse that activates T cells to selectively kill tumor cells while minimizing off-target effects and damage to healthy tissues. Preclinical studies have demonstrated potent in vitro cytotoxicity at picomolar concentrations and efficacy in transgenic mouse models of several cancers, with no observed downregulation of the target surface protein. The drug aims to address challenging cancers such as metastatic castration-resistant prostate cancer (mCRPC) and other solid tumors that are often resistant or less responsive to traditional immunotherapies. Clinical trials are planned for initiation in the second half of 2025[1][3][4][5][6].
