Overview

NRP-1 siRNA is a small interfering RNA (siRNA) therapeutic candidate designed to silence the expression of Neuropilin-1 (NRP-1), specifically the proteoglycan form (PG-NRP1), in metastatic carcinoma-associated fibroblasts (mCAFs). Developed by Environ, this agent aims to modulate the tumor microenvironment by reversing the activated phenotype of CAFs, which are known to promote cancer progression and metastasis. In preclinical studies, knockdown of NRP-1 in mCAFs led to a reduction in activation markers such as alpha-smooth muscle actin (a-SMA), Tenascin C, and Fibronectin, and inhibited the migration and invasion of cancer cells in various solid tumors, including prostate, breast, colorectal, and pancreatic cancers. The siRNA was used to validate NRP-1 as a target for a novel CAF-therapy, leading to the engineering of a small molecule inhibitor for the same pathway.
