Overview

NRP2945 is a synthetic, eleven-amino-acid peptidomimetic analogue of the human CAPS-2 protein, classified as a neural regeneration peptide. It is being developed primarily for neurological disorders with an emphasis on epilepsy syndromes. Preclinical studies have shown that NRP2945 stimulates neural proliferation, migration, and differentiation in the central nervous system without apparent toxicity and with high target specificity. In animal models of epilepsy, it has demonstrated anticonvulsant effects by increasing neuronal survival and differentiation, reducing seizure activity, inhibiting neuronal injury (dark neuron formation), and upregulating GABAA receptor α/β subunits and GAD-65 expression in key brain regions[1][7]. The mechanism of action includes modulation of the CXCR4 receptor as well as neurostimulatory effects[2][5]. Clinical development has focused on absence epilepsy (including drug-resistant forms) and Lennox-Gastaut syndrome; orphan drug status has been granted for Lennox-Gastaut syndrome[2]. The drug crosses the blood-brain barrier rapidly after administration[4].
