NRX-100 is a proprietary, preservative-free formulation of intravenous ketamine developed by NRx Pharmaceuticals for the acute treatment of suicidal crises in depression, including bipolar depression and major depressive disorder. Unlike standard ketamine formulations that contain preservatives such as benzethonium chloride—which have been associated with potential neurotoxicity and cytotoxicity—NRX-100 is specifically designed to avoid these additives, aiming to improve safety for repeated use. The drug leverages the rapid antidepressant effects of ketamine, which are believed to be mediated through antagonism of the brain’s N-methyl D-aspartate (NMDA) receptors. This action modulates glutamate signaling, a pathway implicated in the pathophysiology of depression and suicidality. NRX-100 has received FDA Fast Track designation and is currently under regulatory review following submission of a New Drug Application for use in patients experiencing acute suicidal ideation or behavior[2][4][7][8][9].
