Overview

NS-041 is a proprietary, highly selective small molecule activator of the KCNQ2 and KCNQ3 (Kv7.2/7.3) voltage-gated potassium channels, developed by NeuShen Therapeutics for the treatment of neurological and psychiatric disorders, primarily epilepsy and major depressive disorder. By activating these channels, NS-041 stabilizes neuronal membrane potential and reduces hyperexcitability associated with seizures and mood dysregulation. Preclinical studies demonstrated robust antiseizure efficacy in multiple animal models (MES, PTZ, 6Hz), favorable pharmacokinetic properties suitable for daily oral dosing, and a good safety window with minimal CNS side effects compared to similar compounds. Clinical development has advanced through Phase I trials in healthy volunteers for both epilepsy and major depressive disorder indications; Phase II trials are planned or ongoing for focal epilepsy as an add-on therapy. The drug is positioned as a potential best-in-class agent due to its differentiated efficacy/safety profile[1][2][3][4][5][6][8].
