Overview

NS-863 is an orally bioavailable small molecule prostacyclin receptor (IP receptor) agonist being developed by NS Pharma, a subsidiary of Nippon Shinyaku, for the treatment of pulmonary arterial hypertension (PAH). The drug targets the prostacyclin pathway, which is frequently downregulated in patients with PAH. By selectively binding to and activating the IP receptor on vascular smooth muscle cells, NS-863 promotes potent vasodilation and inhibits the proliferation of smooth muscle cells, thereby aiming to reduce pulmonary vascular resistance and improve exercise capacity. It is currently being evaluated in Phase 2 clinical trials (such as NCT06144125) for patients with Group 1 PAH who are on stable background therapy.
