NS1209 (also known as SPD 502) is a potent, selective, water-soluble small molecule antagonist of the AMPA (α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid) receptor, a subtype of glutamate receptor. It was developed as a neuroprotective agent and antiepileptic drug by NeuroSearch for the treatment of nervous system diseases such as status epilepticus, neuropathic pain, and stroke. The compound demonstrated efficacy in preclinical models for epilepsy and neuropathic pain but failed to show conclusive benefit in clinical trials due to suboptimal study design and insufficient statistical power. Its development was discontinued after Phase II trials for these indications. NeuroSearch ceased R&D operations in 2012; its portfolio was transferred to Teva Pharmaceuticals and Saniona[1][2][3][5].
