NS1619 is a benzimidazolone derivative that acts as a selective activator of large-conductance calcium-activated potassium (BK) channels, also known as KCa1.1 or Maxi-K channels. It is primarily utilized as a pharmacological research tool to study the physiological roles of BK channels in various tissues, including vascular smooth muscle and pulmonary epithelium. In preclinical models of Pseudomonas aeruginosa pneumonia, NS1619 has demonstrated protective effects by reducing pro-inflammatory cytokine production and modulating metabolic pathways, such as those involving itaconate and kynurenine, thereby mitigating lung injury. Although it has been investigated for potential therapeutic applications in hypertension and bladder dysfunction, it is not currently in clinical use and is primarily available as a laboratory reagent.
