Overview

NS19504 is a small-molecule, selective activator of large-conductance calcium-activated potassium (BK, KCa1.1, MaxiK) channels. Originally developed by NeuroSearch, the compound acts as a positive allosteric modulator of the BK channel, left-shifting its voltage activation curve and increasing potassium efflux. In preclinical studies, NS19504 has demonstrated the ability to relax bladder smooth muscle and reduce spontaneous phasic contractions, suggesting potential utility in bladder overactivity and urinary incontinence. Additionally, research has shown that NS19504-mediated BK channel activation exerts neuroprotective and anti-inflammatory effects in astrocytes, indicating potential therapeutic relevance in neuromyelitis optica spectrum disorders (NMOSD).
