Overview

NS2359 is a small molecule, orally active triple monoamine reuptake inhibitor that blocks the neuronal reuptake of serotonin, noradrenaline (norepinephrine), and dopamine. This "triple mode of action" increases extracellular concentrations of these neurotransmitters in the brain and was expected to provide improved efficacy and faster onset compared to existing antidepressants. NS2359 also enhances acetylcholine release in the frontal cortex and hippocampus, which may be relevant for cognitive function. Originally developed as an antidepressant by NeuroSearch and later GlaxoSmithKline (GSK), it was discontinued for major depressive disorder after phase II trials showed lack of efficacy and tolerability. It has also been investigated for attention-deficit hyperactivity disorder (ADHD) and cocaine use disorder; development continues for cocaine-related disorders under Saniona/University of Pennsylvania/National Institute on Drug Abuse[1][3][4][5][7].
