Overview

ns33CAR is an autologous, nanobody-based chimeric antigen receptor (CAR) T-cell therapy targeting CD33, a cell surface antigen highly expressed on myeloid leukemia cells and leukemic stem cells. Developed by Legend Biotech, the therapy utilizes a single-domain antibody (nanobody or VHH) as the antigen-binding domain, which offers potential advantages such as higher binding affinity, improved stability, and reduced immunogenicity compared to traditional scFv-based CAR constructs. The CAR architecture typically incorporates a 4-1BB (CD137) costimulatory domain and a CD3ζ signaling domain to promote T-cell activation, proliferation, and persistence. ns33CAR is primarily being investigated for the treatment of relapsed or refractory acute myeloid leukemia (AML), where it aims to induce deep molecular remissions by specifically eliminating CD33-positive malignant blasts.
