NSC 348884 is a small molecule inhibitor of nucleophosmin (NPM), a multifunctional nucleolar phosphoprotein that is frequently overexpressed or mutated in various human malignancies. The compound specifically targets the N-terminal oligomerization domain of NPM, disrupting the formation of dimers and higher-order oligomers by binding to a defined hydrophobic pocket. This disruption leads to the induction of apoptosis in cancer cells, primarily through the upregulation of p53 via increased phosphorylation at Ser15. Preclinical studies have demonstrated that NSC 348884 inhibits cell proliferation with IC50 values in the low micromolar range across multiple cancer cell lines and can synergize with conventional chemotherapeutic agents like doxorubicin. Currently, it is primarily utilized as a chemical probe in oncology research to study NPM biology and as a lead compound for developing NPM-targeted therapies.
