Overview

NSC-405020 is a small molecule inhibitor that specifically targets Matrix Metalloproteinase-14 (MMP14), also known as Membrane Type 1-Matrix Metalloproteinase (MT1-MMP). Unlike many traditional MMP inhibitors that target the highly conserved catalytic domain, NSC-405020 is designed to interact with the hemopexin domain of MMP14, providing a higher degree of selectivity and avoiding the broad-spectrum toxicity associated with catalytic site inhibitors. In the context of oncology, particularly acute myeloid leukemia (AML), NSC-405020 has been shown to disrupt the supportive crosstalk between mesenchymal stromal cells (MSCs) and leukemic stem cells. By inhibiting MSC-derived MMP14, the compound reduces the secretion of prostaglandin E2 (PGE2), which in turn prevents the activation of the JAK-STAT signaling pathway in leukemia cells. This mechanism leads to decreased leukemia cell proliferation, increased apoptosis, and the reversal of bone marrow-mediated resistance to chemotherapeutic agents like cytarabine (Ara-C).
