Overview

NSC-CRAd-Survivin-pk7 is an investigational cell-based oncolytic virotherapy that uses neural stem cells (NSCs) as carriers to deliver a conditionally replicative adenovirus (CRAd) engineered with the survivin promoter and a polylysine-modified fiber protein (pk7). The survivin promoter restricts viral replication to tumor cells, particularly those overexpressing survivin, such as gliomas and ovarian cancer. The therapy exploits the tumor-tropic properties of NSCs to home in on cancerous tissue, where they release the oncolytic virus. Once inside tumor cells, the virus replicates selectively and lyses them, also stimulating an anti-tumor immune response by exposing new antigens. This approach aims to improve targeting of chemo-resistant or radio-resistant tumors while minimizing toxicity to normal tissues. Clinical trials have shown feasibility and safety in high-grade glioma patients, with ongoing studies for recurrent malignant gliomas and preclinical development for metastatic ovarian cancer[1][2][3][5][8].
