NSC-LIGHT is a genetically engineered stem cell therapy consisting of C17.2 mouse neuronal stem cells (NSCs) retrovirally transduced to express the mouse LIGHT gene (TNFSF14) [1]. Developed by researchers at Kansas State University, NSC-LIGHT leverages the natural tumor-homing ability of NSCs to deliver the immunostimulatory cytokine LIGHT directly to the tumor microenvironment [1]. LIGHT acts as a costimulatory factor that activates lymphoid cells and stimulates T-cell proliferation, promoting tumor cell apoptosis and enhancing anti-tumor immune responses [1]. In preclinical mouse models of highly metastatic mammary tumors, intratumoral administration of NSC-LIGHT significantly attenuated primary tumor growth, reduced metastatic burden, and prolonged survival [1].
