Drug intelligence / Profile preview
NSC232003 is a small molecule inhibitor of Ubiquitin-like with PHD and RING finger domains 1 (UHRF1), an epigenetic regulator that plays a key role in maintaining DNA methylation. NSC232003 binds to the 5-methylcytosine (5mC) binding pocket of the SRA domain of UHRF1, disrupting the interaction between UHRF1 and DNA methyltransferase 1 (DNMT1). This disruption leads to global DNA demethylation and the reactivation of tumor suppressor genes. Preclinical studies have investigated NSC232003 for its anticancer activity in various malignancies, including glioma and bladder cancer, as well as its potential to enhance immunotherapy responsiveness in autosomal dominant polycystic kidney disease (ADPKD) by remodeling the immunosuppressive microenvironment.
