Overview

NSC49L is a small molecule activator of Protein Phosphatase 2A (PP2A), a heterotrimeric serine/threonine phosphatase that functions as a tumor suppressor. PP2A is frequently inactivated in various cancers, contributing to tumor progression and oncogenic signaling. NSC49L was identified as a lead compound for the treatment of colorectal cancer (CRC) and has also been evaluated for its efficacy in melanoma models. It functions by binding to and activating the PP2A holoenzyme, thereby inhibiting downstream oncogenic pathways and inducing apoptosis in cancer cells. NSC49L has served as a structural template for the design of next-generation PP2A activators, such as PPA24 and PPA27, which aim to restore PP2A activity with improved potency in both BRAF-mutant and wild-type melanoma.
