Overview

NSC678917 is a small molecule inhibitor targeting the cytoskeletal scaffolding protein Brain-specific angiogenesis inhibitor 1-associated protein 2 (BAIAP2), also known as IRSp53. Identified through a surface plasmon resonance (SPR) screen by researchers at Georgetown University, the compound is designed to disrupt the BAIAP2 pathway, which is overexpressed in medulloblastoma (MB) and plays a critical role in tumor cell proliferation and migration via interactions with Rho GTPases like CDC42 and RAC1. Preclinical studies indicate that NSC678917 binds directly to BAIAP2 and reduces the viability and invasive potential of medulloblastoma cells, suggesting its potential as a novel therapeutic strategy for pediatric brain tumors.
