Drug intelligence / Profile preview
NSC697923 is a cell-permeable, selective, and covalent small-molecule inhibitor of the ubiquitin-conjugating enzyme E2 N (UBE2N, also known as Ubc13). It specifically targets the catalytic cysteine residue of UBE2N, preventing the formation of the Ubc13-ubiquitin thioester conjugate and thereby disrupting Lysine 63 (K63)-linked polyubiquitination. By blocking K63-linked polyubiquitin chain formation, NSC697923 inhibits downstream signaling pathways, including NF-κB activation and DNA double-strand break repair. Preclinical studies have demonstrated its potential to induce apoptosis and suppress tumor growth in various cancer models, such as neuroblastoma and diffuse large B-cell lymphoma (DLBCL), as well as its utility in studying other K63-ubiquitination-dependent processes like diabetic kidney disease and neurodegenerative protein aggregation.
