NSC750854 is a sulfamated purine analog developed by the National Cancer Institute (NCI) for the treatment of various pediatric cancers. Chemically identified as [3,4-dihydroxy-5-purin-9-yloxolan-2-yl)methyl sulfamate], it represents a novel class of anticancer agents with a profile distinct from traditional purine analogs. Unlike standard purine antimetabolites, it lacks substituents on the purine base and contains a 5'-sulfamate group on its sugar component, which prevents its phosphorylation and subsequent incorporation into DNA or RNA pathways typically affected by purine analogs. Preclinical evaluations conducted by the Pediatric Preclinical Testing Program (PPTP) have shown that NSC750854 exhibits potent cytotoxicity across a wide range of pediatric cell lines and significant tumor growth inhibition in xenograft models of rhabdoid tumors, Wilms tumor, Ewing sarcoma, rhabdomyosarcoma, glioblastoma, and neuroblastoma. Its activity appears to be independent of TP53 status, making it a potential candidate for treating highly resistant pediatric malignancies.
