NSI-566 is a human spinal cord-derived neural stem cell therapy developed as a potential treatment for neurodegenerative and neurological disorders. The therapy consists of a stable line of expanded human neural stem cells derived from the spinal cord tissue of an eight-week-old fetus. Upon transplantation into the central nervous system (typically via intraspinal injection), these cells differentiate into neurons and glia that integrate with host neural networks. They support impaired or damaged motor neurons by forming synaptic connections and secreting neurotrophic factors that promote neuron survival and growth. The primary indications under investigation include amyotrophic lateral sclerosis (ALS), chronic spinal cord injury (cSCI), and paralysis following ischemic stroke or traumatic brain injury. Orphan drug designation has been granted for ALS[1][2][3][5][7][8].
