Overview

NSL-AB-03 is a polyisoprenylated phosphonyl ester inhibitor (PPEI) that acts as a t,t-farnesyl-targeted irreversible inhibitor of polyisoprenylated methylated protein methyl esterase (PMPMEase). PMPMEase is an enzyme involved in the post-translational processing and regulation of monomeric G-proteins, including KRAS, CDC42, RHOA, and RAC1. By inhibiting PMPMEase, NSL-AB-03 disrupts the functional cycle of these G-proteins, leading to the suppression of oncogenic signaling through the MAPK and PI3K/AKT pathways. Preclinical research in pancreatic cancer models has shown that NSL-AB-03 inhibits cell viability and proliferation, induces apoptosis, and causes cytoskeletal disruption. It was developed by researchers at Florida A&M University.
