Overview

NSL-AB-45 is a small molecule polyisoprenylated cysteinyl amide inhibitor (PCAI) being investigated for its potential anticancer properties, particularly in the treatment of pancreatic cancer. Developed by researchers at Florida A&M University, it is a water-soluble analog designed to disrupt the interaction between mutant KRAS and its chaperone proteins. NSL-AB-45 specifically binds to calmodulin, a protein essential for the trafficking and localization of KRAS to the inner surface of the plasma membrane. By inhibiting this interaction, NSL-AB-45 interferes with KRAS-driven oncogenic signaling. In preclinical studies, it has demonstrated significant potency against pancreatic cancer cell lines, including MIA PaCa-2 and PANC-1, with EC50 values in the low micromolar range.
