NSP-1047 is a potent, small molecule inhibitor of MAPK-interacting kinases 1 and 2 (MNK1 and MNK2) developed by Novostar Pharmaceuticals. It functions by blocking the phosphorylation of eukaryotic initiation factor 4E (eIF4E), a key regulator of protein translation that is often dysregulated in cancer. By inhibiting the MNK/eIF4E axis, NSP-1047 suppresses the synthesis of tumorigenic proteins and modulates the expression of immune checkpoint proteins (PD-1, Tim-3, Lag-3) and inflammatory cytokines (IL-6, IL-8, IL-10). Preclinical data in acute myeloid leukemia (AML) models show potent antitumor activity and tumor regression, supported by favorable ADME and pharmacokinetic profiles.
