Overview

NT-0111 is a small-molecule inhibitor targeting Carbonic Anhydrase IX (CAIX), an enzyme that is highly overexpressed in hypoxic tumor environments and plays a critical role in regulating intracellular and extracellular pH. By inhibiting CAIX, NT-0111 disrupts the survival mechanisms of cancer cells in acidic microenvironments, potentially limiting tumor growth, survival, and metastasis. Developed by Nova Paths (also known as Nova Therapeutics) in collaboration with the BC Cancer Foundation, the agent is being investigated for the treatment of various solid and hematological malignancies. Clinical development has included a Phase Ib study in pancreatic ductal adenocarcinoma (PDAC) in combination with gemcitabine, with further Phase II trials planned for 2025-2026.
