NT-0249

NT-0249 is a peripherally restricted, orally bioavailable small molecule inhibitor of the NLRP3 inflammasome. Developed by NodThera, it is designed to treat chronic inflammatory diseases driven by excessive activation of the NLRP3 inflammasome complex. In preclinical models, NT-0249 reduced systemic inflammatory markers and reversed established diet-induced obesity in mice. In Phase 1 clinical studies, it demonstrated significant anti-inflammatory effects in healthy volunteers—reducing key biomarkers such as C-reactive protein (CRP) and fibrinogen—and was well tolerated with a pharmacokinetic profile supporting once-daily dosing. While not as effective for weight loss as semaglutide in animal models, NT-0249 notably reduced cardiovascular inflammation biomarkers associated with heart disease[1][2][7][8].
