NT-1044 is a novel small molecule biguanide and AMPK activator developed by NovaTarg Therapeutics. It was designed to improve upon the anti-tumorigenic properties of metformin by exhibiting higher affinity for organic cation transporters OCT1 (SLC22A1) and OCT3 (SLC22A3), which facilitate cellular entry. In preclinical studies for endometrial cancer, NT-1044 has demonstrated the ability to inhibit cell proliferation, induce G1 cell cycle arrest and apoptosis, and suppress the mTOR pathway (evidenced by decreased S6 phosphorylation). It has also shown efficacy in reducing tumor weight in mouse models of endometrioid endometrial cancer, suggesting potential utility in treating cancers associated with obesity and diabetes.
