NT-112 is an autologous T-cell therapy under development by Neogene Therapeutics (a subsidiary of AstraZeneca) for the treatment of advanced solid tumors, including metastatic non-small cell lung cancer, colorectal adenocarcinoma, pancreatic ductal adenocarcinoma, and endometrial cancer. The therapy consists of autologous T-cells genetically engineered to express an HLA-C*08:02-restricted T cell receptor (TCR) that specifically targets the KRAS G12D mutation. Additionally, these cells are armored by knocking out the gene encoding transforming growth factor beta receptor type 2 (TGFBR2), making them resistant to immunosuppressive effects mediated by TGF-β in the tumor microenvironment. The product is administered via parenteral infusion following lymphodepletion and concurrent recombinant IL-2 support. Preclinical studies have demonstrated high specificity for KRAS G12D mutant cells and favorable safety and efficacy profiles[1][4][6].
