NT-175 is an autologous T cell therapy product genetically engineered to express a T cell receptor (TCR) that is restricted to HLA-A*02:01 and specifically targets the TP53 R175H mutation, a common oncogenic alteration in various solid tumors. The therapy involves editing patient-derived T cells to express this specific TCR, enabling them to recognize and kill tumor cells harboring the TP53 R175H mutation in the context of HLA-A*02:01. Additionally, these engineered T cells are further modified by disrupting the gene encoding transforming growth factor beta receptor 2 (TGFBR2), making them resistant to immunosuppressive effects of TGF-β within the tumor microenvironment. The treatment protocol includes lymphodepleting chemotherapy with fludarabine and cyclophosphamide prior to infusion, followed by administration of recombinant interleukin-2 (rIL-2) post-infusion. NT-175 is being developed for patients with unresectable, advanced or metastatic solid tumors positive for both HLA-A*02:01 and TP53 R175H mutation—including non-small cell lung cancer, colorectal adenocarcinoma, head and neck squamous cell carcinoma, pancreatic adenocarcinoma, ovarian cancer, breast cancer, and other eligible solid tumors[1][5][8].
