NT-391 is a small-molecule inhibitor targeting AXL, a member of the TAM (Tyro3, AXL, MerTK) family of receptor tyrosine kinases. Developed by Nova Paths (also known as Nova Therapeutics), the drug is designed to inhibit AXL signaling, which is frequently upregulated in various cancers and associated with tumor progression, epithelial-mesenchymal transition (EMT), metastasis, and resistance to standard therapies such as checkpoint inhibitors and chemotherapy. NT-391 is currently being evaluated in Phase 2 clinical trials for non-small cell lung cancer (NSCLC) in combination with the PD-1 inhibitor pembrolizumab (Keytruda), and in Phase 1 trials for hematological malignancies, including acute myeloid leukemia (AML) and myelodysplastic syndromes (MDS).
