Overview

**NTB-010** is a nanoparticle albumin-bound (nab) formulation of 17-AAG (17-allylamino-17-demethoxygeldanamycin), a geldanamycin analogue and potent inhibitor of heat shock protein 90 (HSP90). HSP90 is a molecular chaperone critical for the stability and function of oncogenic client proteins, making NTB-010 a targeted small-molecule therapy designed to disrupt cancer cell proliferation and survival. Developed initially by NantBioScience in collaboration with Celgene, it was planned for Phase I trials in hematological malignancies and solid tumors around 2014-2015 to improve the solubility and tolerability of 17-AAG, which had prior development challenges due to hepatotoxicity.[3][7][12]
