Overview

NTDDS-GAD (also known as NG2 GAD) is a gene therapy candidate developed using the Nerve Targeting Drug Delivery System (NTDDS) platform for the treatment of chronic pain, particularly diabetic neuropathic pain. The therapy utilizes a replication-defective herpes simplex virus (HSV) vector to deliver the gene for glutamic acid decarboxylase (GAD) to the dorsal root ganglion (DRG) neurons via intradermal injection. Once the neurons are transduced, they constitutively express GAD, which catalyzes the synthesis of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA) from glutamate. This GABA is then released into the dorsal horn of the spinal cord, where it modulates pain transmission by acting on GABA receptors. Originally developed by Diamyd Medical, the NTDDS platform and the associated GAD programs were divested to Periphagen Holdings in 2012 while in the preclinical stage for the diabetes indication.
