Overview

NTG-101 is a protein-based biologic therapy designed as an injectable treatment for degenerative disc disease (DDD) and related chronic back and neck pain. It consists of a combination of recombinant human transforming growth factor beta 1 (rhTGF-β1) and connective tissue growth factor (rhCTGF), delivered via intradiscal injection. The mechanism of action involves anti-inflammatory, anti-catabolic, and pro-anabolic effects within the intervertebral disc, suppressing pro-inflammatory cytokines (such as IL-6, IL-8), reducing pain-related neurotrophins (NGFr, BDNF, TrkB, CALCRL), halting disc degeneration, and promoting repair by stimulating extracellular matrix proteins like aggrecan and collagen II. Developed by Notogen for regenerative therapy in spinal disorders, NTG-101 is currently in preclinical development[1][3][4][5][6].
