NTS-521 is a small molecule agonist of the Tropomyosin receptor kinase B (TrkB), which is the primary high-affinity receptor for brain-derived neurotrophic factor (BDNF). Developed by Neurotrauma Sciences under a licensing agreement with Emory University, NTS-521 is designed to mimic the neuroprotective and neuroregenerative effects of BDNF while overcoming its inherent pharmacological limitations, such as poor blood-brain barrier penetration and a short half-life. By activating the TrkB signaling pathway, the compound aims to promote neuronal survival, enhance synaptic plasticity, and facilitate repair following central nervous system injury. NTS-521 is currently in preclinical development, with primary research focusing on its potential to treat traumatic brain injury (TBI) and ischemic stroke.
