NTS071 is an oral small molecule allosteric reactivator specifically designed to target the p53 Y220C mutation, a cancer-associated mutant form of the tumor suppressor protein p53. Developed by Nutshell Therapeutics using their proprietary AI-driven ALLOSTAR™ platform, NTS071 selectively binds to and stabilizes the p53 Y220C mutant protein. This binding improves the thermal stability of the mutant protein, restores its DNA-binding and transcriptional activity, and reactivates its tumor-suppressing function. Preclinical studies have shown that NTS071 exhibits high potency (picomolar-level biochemical activity), superior metabolic stability across species, lower in vivo clearance rates, higher oral exposure compared to competitor compounds like PC14586 (rezatapopt), and a favorable safety profile with reduced risk for drug-drug interactions due to minimal CYP3A4 inhibition. The drug is being developed as a potential best-in-class therapy for solid tumors harboring TP53 Y220C mutations[1][2][4][10].
