NTX-101 is a first-in-class small molecule eye drop developed for the treatment of glaucoma. Its mechanism of action involves inhibition of 11β-HSD1 (Corticosteroid 11-beta-dehydrogenase isozyme 1), which leads to both lowering intraocular pressure and providing direct protection to the optic nerve. By targeting this enzyme, NTX-101 modulates cortisol levels in ocular tissues, offering neuroprotective effects beyond traditional therapies that primarily focus on reducing intraocular pressure. The drug is being developed by Pinotbio and has reached Phase 2 clinical development for glaucoma[1][2][3]. There are also early-stage investigations into its use for other retinal neovascular conditions such as wet age-related macular degeneration (AMD) and diabetic retinopathy[4].
