Overview

NTX-1088 is a first-in-class, highly potent monoclonal antibody that targets the poliovirus receptor (PVR, also known as CD155), a transmembrane protein expressed on cancer cells. Developed for the treatment of solid tumors, NTX-1088 works by binding to PVR with high affinity and blocking its interactions with immune checkpoint receptors TIGIT and CD96, thereby preventing their inhibitory signaling. Additionally, it restores DNAM1 (CD226) function on T and NK cells by preventing its internalization and downregulation caused by tumor cells. This triple mechanism of action aims to reinstate antitumor immune activity and enhance immune cell activation against tumors. NTX-1088 is being developed by Nectin Therapeutics and is currently in Phase I clinical trials as monotherapy and in combination with pembrolizumab for patients with locally advanced or metastatic solid tumors[1][2][3][4][5][6][8].
