NTX-250 is a novel, multimodal mRNA-based immunotherapy developed by Nutcracker Therapeutics for the treatment of tumors driven by human papillomavirus type 16 (HPV16), including cervical intraepithelial neoplasia (CIN) and cervical cancer. The therapy consists of three synergistic mRNA molecules encoding HPV16 E6/E7 antigens, interleukin-12 (IL-12), and LIGHT (TNFSF14). Delivered intratumorally, these components are designed to induce robust antigen-specific T cell responses, modulate the tumor microenvironment to enhance immune cell infiltration and activity, and reduce immunosuppression. Preclinical studies have demonstrated complete regression of large HPV-driven tumors in murine models with long-term survival and immune memory against re-challenge. Translational potential has also been shown in non-human primates with strong HPV-specific immune responses[2][5][6][7][8][9].
