NTX-301 + platinum-based chemotherapy is an investigational combination therapy consisting of NTX-301, an orally bioavailable hypomethylating agent (5-aza-4′-thio-2′-deoxycytidine), and a platinum-based chemotherapeutic (such as carboplatin or cisplatin). NTX-301 is a potent inhibitor of DNA methyltransferase 1 (DNMT1) that induces epigenetic modulation by DNA hypomethylation, leading to reactivation of tumor suppressor genes, disruption of cancer cell lipid metabolism, targeting of cancer stem cells, induction of ferroptosis, and anti-tumor activity. In preclinical and early clinical studies, this combination is being developed to address platinum-resistant or refractory advanced ovarian cancer, bladder cancer, and select other solid tumors by combining the epigenetic reprogramming of NTX-301 with DNA-damaging effects of platinum agents[2][3][5][7][8][9].
