Overview

NTX-352 is a clinical-stage combination therapy program developed by NeoTX Therapeutics. It centers on naptumomab estafenatox (NAP), a tumor-targeted superantigen (TTS) biologic. NAP consists of a Fab fragment targeting the 5T4 oncofetal antigen fused to an engineered version of the staphylococcal enterotoxin A (SEA/E-120) superantigen. This superantigen component binds to the T-cell receptor (TCR) of a large subset of T cells, bypassing MHC restriction to induce a potent localized immune response within the tumor microenvironment. NTX-352 involves combining NAP with various agents, including the PD-L1 inhibitor durvalumab (in partnership with AstraZeneca), the chemotherapy docetaxel, or the personalized neoantigen peptide vaccine NeoPep GT. These combinations aim to enhance tumor-directed immune activation or leverage chemotherapy-mediated tumor killing.
