NTX-353 is a preclinical-stage tumor-targeted superantigen (TTS) biologic being developed by NeoTX Therapeutics in collaboration with the Dana-Farber Cancer Institute for the treatment of glioblastoma multiforme (GBM). The TTS platform utilizes a fusion protein comprising a tumor-targeting moiety and a bacterial superantigen. This approach is designed to induce a potent T-cell mediated immune response directly within the tumor microenvironment by bypassing the requirement for specific antigen presentation. By coating tumor cells with superantigens, NTX-353 aims to activate a large population of T cells to recognize and destroy the glioblastoma cells, potentially overcoming the highly immunosuppressive nature of central nervous system tumors.
