NTX-354 is a preclinical-stage therapeutic candidate developed by NeoTX Therapeutics that combines a tumor-targeted superantigen (TTS) with CAR-T cell therapy. The TTS component is a fusion protein designed to bind to specific antigens on the surface of tumor cells and simultaneously present a bacterial superantigen. This superantigen acts as a potent immune stimulant, recruiting and activating T cells—including the co-administered CAR-T cells—directly to the tumor microenvironment. By bridging the engineered CAR-T cells to the tumor via the superantigen-MHC/TCR interaction, NTX-354 aims to enhance the infiltration, persistence, and cytotoxic efficacy of CAR-T therapy, particularly in hematologic cancers.
