Overview

NTX-470 is a stabilized, cyclized peptide therapeutic being developed for the treatment of Alzheimer's disease. It was discovered by Neuro-Bio Ltd, a biotechnology company in which Addex Therapeutics holds a financial interest through a 20% share in a royalty and milestone LLC. The drug is designed to block the effects of T14, a toxic 14-amino acid peptide derived from the C-terminus of acetylcholinesterase (AChE). T14 is hypothesized to drive neurodegeneration by binding to an allosteric site on the alpha-7 nicotinic acetylcholine receptor (α7nAChR), leading to pathological calcium over-entry and subsequent neuronal death. NTX-470 acts as a T14 antagonist, competing for the same binding site on the α7nAChR to prevent this neurotoxic cascade. It is intended for intranasal administration to facilitate direct delivery to the brain and is currently in preclinical development.
