Overview

NTX-532 is a small molecule positive allosteric modulator (PAM) of the GABAB receptor, developed by Addex Therapeutics. It is part of a portfolio of GABAB PAM assets, which also includes NTX-470 and NTX-618, that are subject to a royalty and milestone agreement with 20% Royalty/Milestone LLC. The GABAB PAM program is primarily targeted at treating substance use disorders (including alcohol and opioid use disorders) and chronic cough. By acting as a PAM, NTX-532 enhances the inhibitory effects of the endogenous neurotransmitter GABA at the GABAB receptor without the significant side effects, such as sedation and muscle weakness, typically associated with orthosteric GABAB agonists like baclofen. Addex Therapeutics regained the global rights to this program from Indivior in early 2024.
