NTX-618 is a small molecule positive allosteric modulator (PAM) of the gamma-aminobutyric acid type B (GABAB) receptor. Originally developed by Addex Therapeutics, the compound is part of a portfolio of GABAB PAM assets (including NTX-470 and NTX-532) that was transitioned to Neuro-Therapeutics LLC. Addex Therapeutics retains a 20% interest in future milestones and royalties from these programs through a specialized entity often referred to as 20% Royalty/Milestone LLC. As a GABAB PAM, NTX-618 is designed to selectively enhance the inhibitory signaling of the GABAB receptor in the presence of endogenous GABA, potentially providing therapeutic effects for central nervous system (CNS) and peripheral disorders such as chronic cough and substance use disorders, while minimizing the side effects typically associated with orthosteric GABAB agonists.
