NTX-748 is an orally available small molecule inhibitor of CD38 (ADP-ribosyl cyclase/cyclic ADP-ribose hydrolase 1), an enzyme that consumes NAD+ and is ubiquitously expressed on immune cells. Developed by Juvenescence through its portfolio company Napa Therapeutics, NTX-748 is being investigated for the treatment of rheumatoid arthritis, where CD38 expression is known to increase. By inhibiting the enzymatic activity of CD38, the drug aims to increase cellular NAD+ levels and modulate inflammatory processes. Preclinical studies in collagen-induced arthritis mouse models have demonstrated that NTX-748 provides dose-dependent beneficial effects, reducing clinical arthritis scores and disease incidence without significant toxicity.
